Are you tying to connect toa database? If so, make sure to check all your permissions on the DB and its containing folders

Where are you testing on a live server or a testing server? Are using a dns-less or a dns connection.
Also, check the User permissions on your folder where the Db is located and make user user can Write to it. I guess you did that, oh well check again.

Please check if ya have given appropriate permission on the folder where your database resides. make sure ya give permission to the "Internet Guess Account". I believe this should solve your problem.
